Staurosporine induces lamellipodial widening in locomoting fish keratocytes by abolishing the gradient from radial extension of leading edge

چکیده
Fish epidermal keratocytes locomote along surfaces without overall cell size or shape changes, as kinematically described by the graded radial extension (GRE) model. We found that the cell size increased during locomotion after the addition of a low dose of staurosporine or K-252a, broad-spectrum protein-serine/threonine kinase inhibitors. Quantitative shape analysis showed that the cell size increase resulted from an increase in lamellipodial width, the maximal length perpendicular to the direction of the cell locomotion, whereas the lamellipodial length, along the locomotion direction, remained constant. Importantly, the gradient of radial extension in the leading edge disappeared during lamellipodial width increase. These results suggest that a special mechanism for producing graded radial extension of lamellipodia exists to keep cell size constant, and that a protein-serine/threonine kinase plays an important role in regulating this mechanism.

Analysis of the Actin–Myosin II System in Fish Epidermal Keratocytes: Mechanism of Cell Body Translocation
While the protrusive event of cell locomotion is thought to be driven by actin polymerization, the mechanism of forward translocation of the cell body is unclear. To elucidate the mechanism of cell body translocation, we analyzed the supramolecular organization of the actin-myosin II system and the dynamics of myosin II in fish epidermal keratocytes. In lamellipodia, long actin filaments formed...
متن کاملThe composition and dynamics of cell-substratum adhesions in locomoting fish keratocytes.
Close contacts are the predominant type of cell-substratum adhesion in rapidly moving cells yet little is known about their composition and dynamics. To address these issues we have attempted to identify the molecular components of close contacts formed in rapidly moving fish epithelial keratocytes. In addition we have utilized the simple shape of keratocytes to explore the relationship between...
متن کاملSeparation of Propulsive and Adhesive Traction Stresses in Locomoting Keratocytes
Strong, actomyosin-dependent, pinching tractions in steadily locomoting (gliding) fish keratocytes revealed by traction imaging present a paradox, since only forces perpendicular to the direction of locomotion are apparent, leaving the actual propulsive forces unresolved. When keratocytes become transiently "stuck" by their trailing edge and adopt a fibroblast-like morphology, the tractions opp...
متن کاملTraction forces generated by locomoting keratocytes
Traction forces produced by moving fibroblasts have been observed as distortions in flexible substrata including wrinkling of thin, silicone rubber films. Traction forces generated by fibroblast lamellae were thought to represent the forces required to move the cell forwards. However, traction forces could not be detected with faster moving cell types such as leukocytes and growth cones (Harris...

We have previously reported that rearward migration of surface particles on slowly moving cells is not driven by membrane flow (Sheetz, M. P., S. Turney, H. Qian, and E. L. Elson. 1989. Nature (Lond.). 340:284-288) and recent photobleaching measurements have ruled out any rapid rearward lipid flow (Lee, J., M. Gustafsson, D. E. Magnussen, and K. Jacobson. 1990. Science (Wash.
